چکیده
An in vitro RNA-synthesizing system was used to study the effects of S-adenosyl-L-homocysteine, S-adenosyl-L-methionine, and adenosine on the methylation and synthesis of single-stranded RNA by two different cytoplasmic polyhedrosis viruses.

CPV, a stable and symmetrical machine for mRNA synthesis.
The structure of cytoplasmic polyhedrosis virus (CPV), an insect pathogen from the Reoviridae family of double-strand RNA viruses, has been determined at 8 A by electron cryomicroscopy and image reconstruction. It provides new information about the functions of these viral particles as stable machines for mRNA synthesis.
متن کاملDiscovery of m7G-cap in eukaryotic mRNAs
Terminal structure analysis of an insect cytoplasmic polyhedrosis virus (CPV) genome RNA in the early 1970s at the National Institute of Genetics in Japan yielded a 2'-O-methylated nucleotide in the 5' end of double-stranded RNA genome. This finding prompted me to add S-adenosyl-L-methionine, a natural methylation donor, to the in vitro transcription reaction of viruses that contain RNA polymer...
متن کاملA putative ATPase mediates RNA transcription and capping in a dsRNA virus
mRNA transcription in dsRNA viruses is a highly regulated process but the mechanism of this regulation is not known. Here, by nucleoside triphosphatase (NTPase) assay and comparisons of six high-resolution (2.9-3.1 Å) cryo-electron microscopy structures of cytoplasmic polyhedrosis virus with bound ligands, we show that the large sub-domain of the guanylyltransferase (GTase) domain of the turret...
متن کاملS-Inosyl-L-Homocysteine Hydrolase, a Novel Enzyme Involved in S-Adenosyl-L-Methionine Recycling.
UNLABELLED S-Adenosyl-L-homocysteine, the product of S-adenosyl-L-methionine (SAM) methyltransferases, is known to be a strong feedback inhibitor of these enzymes. A hydrolase specific for S-adenosyl-L-homocysteine produces L-homocysteine, which is remethylated to methionine and can be used to regenerate SAM. Here, we show that the annotated S-adenosyl-L-homocysteine hydrolase in Methanocaldoco...
متن کاملS-adenosyl-L-homocysteine hydrolase, key enzyme of methylation metabolism, regulates phosphatidylcholine synthesis and triacylglycerol homeostasis in yeast: implications for homocysteine as a risk factor of atherosclerosis.
In eukaryotes, S-adenosyl-L-homocysteine hydrolase (Sah1) offers a single way for degradation of S-adenosyl-L-homocysteine, a product and potent competitive inhibitor of S-adenosyl-L-methionine (AdoMet)-dependent methyltransferases. De novo phosphatidylcholine (PC) synthesis requires three AdoMet-dependent methylation steps. Here we show that down-regulation of SAH1 expression in yeast leads to...
